Is there a way to hotkey screen rotation? Exactly like how (for example) Fullscreen can be a hotkey, asking for games that require rotating the DS in-game.

Ah, you mean like Pokemon Ranger: Guardian Signs? While MelonDS doesn't have a hotkey for that (though I guess Arisotura could add this in a future version), you can rotate the screen through the menu. You'll have to make do with that for now.
